Isotypecontrolsareatypeofnegativecontroldesignedtomeasurethelevelofnon-specificbackgroundsignalcausedbyprimaryantibodies, baseduponthetissuetypeofthesample. Non-specificbindingofprimaryantibodiesaretypicallycausedbythefollowingreasons
Usually,thebackgroundsignalistheresultofimmunoglobulinsbindingnon-specificallytoFcreceptorspresentonthecellsurface.Forexample, antibodiesraisedinmice,particularlythoseoftheIgG2aisotype,willbindstronglytosomehumanleukocytesregardlessofthetestantibody specificity.InthiscaseyouwouldneedamouseIgG2aisotypecontrolforusewithhumancellsortissues.Isotypecontrolsaremostcommonly usedinflowcytometryexperiments,butmayalsobeusedinimmunohistochemistry.
SelectinganIsotypeControl
Typically,anisotypecontrolismatchedtothehostspeciesandisotypeofyourspecificprimaryantibody,the heavychain (IgA,IgG,IgD,IgE,orIgM), subclass and lightchain (kappa,lamBDa)classoftheprimaryantibody; thesameFluorochrome(PE,APC,etc.),andhavethesameF:Pratio.F:Pisameasurementofhowmanyfluorescentmoleculesare presentoneachantibody.
Whenusingdirectlylabeledprimaryantibodies,itisalsonecessarytomakesurethattheisotypecontrolisconjugatedtothesamefluorochrome orlabelasthetestantibody.Itisnotsufficienttouseonewithaspectrallysimilarfluorochrome.
Non-specificBinding
Toavoidnon-specificbinding,youalsoneedtoblockFcreceptorsoncelltypes,suchasspleencells,withFcRblockingreagents.Isotypecontrolsshouldbeusedatthesameconcentration/dilutionastheprimaryantibody.Isotypecontrolsaremainlyusedtodetermineifthestainingisreal.Apartfromisotypecontrols,unstainedcellsshouldalwaysbeincludedinthe experimentalset-uptomonitorautofluorescence.
ApplicationsforIsotypeControls
Althoughisotypecontrolsaremainlyusedinflowcytometry,theycanbeusedasstandardblockingagentsandproteincoatingagentsforotherapplicationsincluding immunofluorescence,immunocytochemistry,westernblotting,andELISA.
Isotypecontrolsareusedtodiscriminatenon-specificbindingofmouseIgG2bprimarymonoclonalantibodiestohumancellsduetoFcrecepterbindingorotherprotein-proteininteractions.

FlowCytometry:Use10uloftheoptimalworkingdilutiontolabel10e6cellsin100ul.Suitableforwholeblood,Ficoll-separatedpreparations.Optimaldilutionstobedeterminedbyresearcher.

StorageandStABIlity
Storeproductat4°Ciftobeusedimmediatelywithintwoweeks.Forlong-termstorage,aliquottoavoidrepeatedfreezingandthawingandstoreat-20°C.Aliquotsarestableat-20°Cfor12monthsafterreceipt.Diluterequiredamountonlypriortoimmediateuse.Furtherdilutionscanbemadeinassaybuffer.Caution:FITCconjugatesaresensitivetolight.Formaximumrecoveryofproduct,centrifugetheoriginalvialafterthawingandpriortoremovingthecap.
