ProperprimerdesigniscriticalforsuccessfulPCRandtransformationsteps.Primersforthegeneofinterestshouldcontaintherestrictionsiteaswellasatleast10nucleotidesofsequencehomologoustothevectorsequence.Usingtwodifferentrestrictionenzymeswillallowdirectionalcloningoftheinsert.Note:Oncepurified,thedigestedvectorcanbeusedwithmultipleinsertsiftheprimersforinsertamplificationaredesignedcorrectly.
