As a member of B7 family, B7-H4 (VTCN1) plays a critical role in the regulation of antigen-specific immune responses. B7-H4 is a co-stimulatory protein expressed in activated T cells and is involved in the negative regulation of cell-mediated immunity in peripheral tissues. B7-H4 ligation to its receptor BTLA T-lymphocytes results in inhibition of T-cell activation, cytokine secretion, and the development of cytotoxicity.
