HepatitisCvirus(HCV)isanenvelope,singlestrandedpositivesenseRNA(9.5kb)virusbelongingtothefamilyofFlaviviridae.SixmajorgenotypesandseriesofsubtypesofHCVhavebeenidentified.Isolatedin1989,HCVisnowrecognizedasthemajorcausefortransfusionassociatednon-A,non-Bhepatitis.Thediseaseischaracterizedwithacuteandchronicformalthoughmorethan50%oftheinfectedindividualsdevelopsevere,lifethreateningchronichepatitiswithlivercirrhosisandhepatocellularcarcinomas.Sincetheintroductionin1990ofanti-HCVscreeningofblooddonations,theincidenceofthisinfectionintransfusionrecipientshasbeensignificantlyreduced.

ThisBioAssay™kitisanenzyme-linkedimmunosorbentassayforqualitativedetectionofantibodiestohepatitisCvirusinhumanserumorplasma.ItisintendedforscreeningblooddonorsanddiagnosingpatientsrelatedtoinfectionwithhepatitisCvirus.

TestPrinciple
Thiskitisatwo-stepincubationenzymeimmunoassay,whichusespolystyrenemicrowellstripspre-coatedwithrecombinantHCVantigensexpressedinE.coli(recombinantCoreandNS3/4/5).Serumorplasmasampleisaddedtogetherwithbiotin-conjugatedHCVantigens.Duringthefirstincubationstep,thespecificHCVantibodies,ifpresent,willbecapturedinsidethewellsasadoubleantigencomplexcomprisingofthecoated,andthebiotin-conjugatedHCVantigens.Themicrowellsarethenwashedtoremoveunboundserumproteins.Duringthesecondincubationstep,thecapturedHCVantibodiesaredetectedbyaddingofHRP-Conjugate.Themicrowellsarethenwashedtoremoveunboundconjugate,andChromogensolutionsareaddedtothewells.InwellspositiveforHCVantibodies,thecolorlessChromogensarehydrolyzedbytheboundHRPconjugatetoabluecoloredproduct.Thebluecolorturnsyellowafterstoppingthereactionwithsulfuricacid.Theamountofcolorintensitycanbemeasuredandisproportionaltotheamountofantibodiescapturedinthewells,andtothesamplerespectively.Wellscontainingsamplesnegativeforanti-HCVremaincolorless.
