Thisassayemploysthesandwichenzymeimmunoassaytechnique.Anti-Infliximabiscoatedontoa96-wellmicroplate.Calibrator,qualitycontrolsamples(ifdesired)andtestsamplesarePipettedintotheappropriatewells.Infliximabpresentinbiologicalmatricesisboundbytheimmobilizedanti-Infliximabantibody.Afterwashingawayanyunboundsubstances,enzyme-linkedanti-Infliximabantibodyisaddedtothewells.ThisantibodyisdevelopedandpurifiedspecificallyagainsttruncatedRemicade®(domainresidinginFcportionoftheRemicade®molecule).Theplateiswashedtoremoveanyunboundantibody-enzymereagentandasubstratesolutionisaddedtothewellsforcolordevelopment.Thecolordevelopmentisstoppedandtheintensityofthecolorismeasured.ThecolorintensityisproportionaltotheamountofInfliximabpresentintestsamples.
