ThisBioAssay™ELISAkitusesSandwich-ELISAasthemethod.ThemicroELISAplateprovidedinthiskithasbeenpre-coatedwithanantibodyspecifictohumanIgA(ImmumoglobulinA).StandardsorsamplesareaddedtotheappropriatemicroELISAplatewellsandcombinedwiththespecificantibody.ThenabiotinylateddetectionantibodyspecificforhumanIgA(ImmumoglobulinA)andAvidin-HorserADIshPeroxidase(HRP)conjugateareaddedtoeachmicroplatewellsuccessivelyandincubated.Freecomponentsarewashedaway.Thesubstratesolutionisaddedtoeachwell.OnlythosewellsthatcontainhumanIgA(ImmumoglobulinA),biotinylateddetectionantibodyandAvidin-HRPconjugatewillappearblueincolor.Theenzyme-substratereactionisterminatedbyaddingStopSolutionandthecolorturnsyellow.Theopticaldensity(OD)ismeasuredspectrophotometricallyatawavelengthof450nm±2nm.TheODvalueisproportionaltotheconcentrationofhumanIgA(ImmumoglobulinA).TheconcentrationofhumanIgA(ImmumoglobulinA)insamplescanbecalculatedbycomparingtheODofthesamplestothestandardcurve.
