ThisBioAssay™ELISAkitusesSandwich-ELISAasthemethod.ThemicroELISAplateprovidedinthiskithasbeenpre-coatedwithanantibodyspecifictohumanEXT1(Exostoses1).StandardsorsamplesareaddedtotheappropriatemicroELISAplatewellsandcombinedwiththespecificantibody.ThenabiotinylateddetectionantibodyspecificforhumanEXT1(Exostoses1)andAvidin-HorserADIshPeroxidase(HRP)conjugateareaddedtoeachmicroplatewellsuccessivelyandincubated.Freecomponentsarewashedaway.Thesubstratesolutionisaddedtoeachwell.OnlythosewellsthatcontainhumanEXT1(Exostoses1),biotinylateddetectionantibodyandAvidin-HRPconjugatewillappearblueincolor.Theenzyme-substratereactionisterminatedbyaddingStopSolutionandthecolorturnsyellow.Theopticaldensity(OD)ismeasuredspectrophotometricallyatawavelengthof450nm±2nm.TheODvalueisproportionaltotheconcentrationofhumanEXT1(Exostoses1).TheconcentrationofhumanEXT1(Exostoses1)insamplescanbecalculatedbycomparingtheODofthesamplestothestandardcurve.
