ThisBioAssay™ELISAkitusesSandwich-ELISAasthemethod.ThemicroELISAplateprovidedinthiskithasbeenpre-coatedwithanantibodyspecifictohumanCS(CitrateSynthase).StandardsorsamplesareaddedtotheappropriatemicroELISAplatewellsandcombinedwiththespecificantibody.ThenabiotinylateddetectionantibodyspecificforhumanCS(CitrateSynthase)andAvidin-HorserADIshPeroxidase(HRP)conjugateareaddedtoeachmicroplatewellsuccessivelyandincubated.Freecomponentsarewashedaway.Thesubstratesolutionisaddedtoeachwell.OnlythosewellsthatcontainhumanCS(CitrateSynthase),biotinylateddetectionantibodyandAvidin-HRPconjugatewillappearblueincolor.Theenzyme-substratereactionisterminatedbyaddingStopSolutionandthecolorturnsyellow.Theopticaldensity(OD)ismeasuredspectrophotometricallyatawavelengthof450nm±2nm.TheODvalueisproportionaltotheconcentrationofhumanCS(CitrateSynthase).TheconcentrationofhumanCS(CitrateSynthase)insamplescanbecalculatedbycomparingtheODofthesamplestothestandardcurve.
