PhospholipaseCZeta1(PLCz1)BioAssay™ELISAKit(Human)isaSandwichenzymeimmunoassay.Themicrotiterplateprovidedinthiskithasbeenpre-coatedwithanantibodyspecifictoPhospholipaseCZeta1(PLCz1).Standardsorsamplesarethenaddedtotheappropriatemicrotiterplatewellswithabiotin-conjugatedantibodyspecifictoPhospholipaseCZeta1(PLCz1).Next,AvidinconjugatedtoHorserADIshPeroxidase(HRP)isaddedtoeachmicroplatewellandincubated.AfterTMBsubstratesolutionisadded,onlythosewellsthatcontainPhospholipaseCZeta1(PLCz1),biotin-conjugatedantibodyandenzyme-conjugatedAvidinwillexhibitachangeincolor.Theenzyme-substratereactionisterminatedbytheadditionofsulphuricacidsolutionandthecolorchangeismeasuredspectrophotometricallyatawavelengthof450nm±10nm.TheconcentrationofPhospholipaseCZeta1(PLCz1)inthesamplesisthendeterminedbycomparingtheO.D.ofthesamplestothestandardcurve.
