Claudin5(CLDN5)BioAssay™ELISAKit(Mouse)isaSandwichenzymeimmunoassay.Themicrotiterplateprovidedinthiskithasbeenpre-coatedwithanantibodyspecifictoClaudin5(CLDN5).Standardsorsamplesarethenaddedtotheappropriatemicrotiterplatewellswithabiotin-conjugatedantibodyspecifictoClaudin5(CLDN5).Next,AvidinconjugatedtoHorserADIshPeroxidase(HRP)isaddedtoeachmicroplatewellandincubated.AfterTMBsubstratesolutionisadded,onlythosewellsthatcontainClaudin5(CLDN5),biotin-conjugatedantibodyandenzyme-conjugatedAvidinwillexhibitachangeincolor.Theenzyme-substratereactionisterminatedbytheadditionofsulphuricacidsolutionandthecolorchangeismeasuredspectrophotometricallyatawavelengthof450nm±10nm.TheconcentrationofClaudin5(CLDN5)inthesamplesisthendeterminedbycomparingtheO.D.ofthesamplestothestandardcurve.
