PenicillinwasaccidentallydetectedbyAlexanderFlemingin1929.ThedrugbelongstothemycotoxinsandisgeneratedbythemouldPenicilliumchrysogenum.Penicillinasanantibioticispreferentiallyusedinthetreatmentofgram-positivebacteria,bothforhumansandanimals.OfallillegallyadmiNISTereddrugs,antibioticsaremostfrequentlyusedinproductivelivestock.Contaminationsinfoodormilkareingestedbyhumans,andcanleadtosevereinfectionsbypathogengermswhichbecameresistantagainstpenicillin,ortoallergies.Theallergicreactionsappearwithdifferentseverity,dependentondoseandindividualdisposition,andshowingsymptomsfromurticariatoanaphylacticshock.Duringroutinetestingofmilksamplesforantibiotics,inmorethan90%ofthepositivecases,betalactampreparationsorpenicillinsaredetected.

TestPrinciple
TheUnitedStatesBiologicalPenicillinquantitativetestisbasedontheprincipleoftheenzymelinkedimmunosorbentassay.Apenicillinconjugateisboundonthesurfaceofamicrotiterplate.Penicillincontainingsamplesorstandardsandanantibodydirectedagainstpenicillinaregivenintothewellsofthemicrotiterplate.Immobilizedandfreepenicillincompetefortheantibodybindingsites.Afteronehourincubationatroomtemperature,thewellsarewashedwithdilutedwashingsolutiontoremoveunboundmaterial.Aperoxidaseconjugatedirectedagainstthepenicillinantibodyisgivenintothewellsandafteranotherhourincubation,theplateiswashedagain.Thenasubstratesolutionisaddedandincubatedfor20minutes,resultinginthedevelopmentofabluecolor.Thecolordevelopmentisinhibitedbytheadditionofastopsolution,andthecolorturnsyellow.Theyellowcolorismeasuredphotometricallyat450nm.Theconcentrationofpenicillinisindirectlyproportionaltothecolorintensityofthetestsample.
