Calnexin,anabundant~90kDintegralproteinoftheendoplasmicreticulum,isalsoreferredtoasIP90,p88andp90(1).Itconsistsofalarge50kDN-terminalcalcium-bindingluminaldomain,asingletransmembranehelixandashortacidiccytoplasmictail(2,3).UnlikeitsERcounterpartswhichhaveaKDELsequenceontheirC-terminustoensureERretention(4),calnexinhaspositivelychargedcytosolicresiduesthatdothesamething(3).MostERproteinsactasmolecularchaperonesandparticipateintheproperfoldingofpolypeptidesandtheirassemblyintomulti-subunitproteins.Calnexintogetherwithcalreticulin,playsakeyroleinglycoproteinfoldinganditscontrolwithintheER,byinteractingwithfoldingintermediatesviatheirmono-glycosylatedglycans(5,6).CalnexinhasalsobeenshowntoassociatewiththemajorhistocompatibilitycomplexclassIheavychains,partialcomplexesoftheTcellreceptorandBcellmembraneimmunoglobulin(7).
