Ferritinisimportantinironhomeostasis.Itisthemainiron-storageprotein,composedoftwopartiallyhomologoussubunits,heavyandlightchains.Ferritinmoleculesincellscontaininghighlevelsofirontendtoberichinlightchains,andmayhavealong-termstoragefunction,whereasheavy-richferritinsaremoreactiveinironmetabolism.Mutationsintheferritingenecausethehereditaryhyperferritinemia-cataractsyndromeandneuroferritinopathy,associatedwithinflammation.Elevatedlevelsofferritinarereportedascharacteristicofadult-onsetStill'sdiseaseandhemophagocyticsyndrome,alsoassociatedwithinflammation.MICROFILamentsandincreasedlevelsofexogenousferritinareexcreteddirectlyintothebilebywayofasecondmicrofilament-independent,chloroquine-insensitivepathway,supportingapossIBLephysiologicalmechanismforthereleaseofferritinfromtheliver.

StorageandStABIlity

Maybestoredat4°Cforshort-termonly.Forlong-termstorageandtoavoidrepeatedfreezingandthawing,aliquotandstoreat-20°C.Aliquotsarestablefor12monthsafterreceiptat-20°C.Formaximumrecoveryofproduct,centrifugetheoriginalvialafterthawingandpriortoremovingthecap.Furtherdilutionscanbemadeinassaybuffer.FITCconjugatesaresensitivetolight.
