WestNileVirus(WNV)isamemberoftheFlaviviridae,aplus-strandedvirusfamilythatincludesSt.Louisencephalitisvirus,yellowfevervirus,andDenguevirus.WNVwasinitiallyisolatedin1937intheWestNileregionofUgandaandhasbecomeprevalentinAfrica,Asia,andEurope.IthasrapidlyspreadacrosstheUnitedStateswithcasesbeingobservedineverycontinentalstate.Virusparticlesconsistofadensecoremadeupofthecore/capsidproteinencapsulatingtheRNAgenomesurroundedbyamembraneenvelopeembeddedwithenvelopeandmatrixproteins.WhiletheviralcoreproteinisthoughttocontributetotheWNV-associatedinflammationviaapoptosisinducedthoughthecaspase-9pathway,thehighlyglycosylatedenvelopeproteinplaysamajorroleforWNVentryintotargetcellsasthisentrycanbeinhibitedbyusingarecombinantdomainIIIfromtheenvelopeglycoprotein.TheWNVreceptorhasrecentlybeenidentifiedasalphavbeta3integrin.
