In1990anoncogene,v-mpl,wasidentifiedfromthemurinemyeloproliferativeleukemiavirusthatwascapableofimmortalizingbonemarrowhematopoieticcellsfromdifferentlineages.In1992thehumanhomologue,named,c-mpl,wascloned.Sequencedatarevealedthatc-mplencodedaproteinthatwashomologouswithmembersofthehematopoieticreceptorsuperfamily.Presenceofanti-senseoligodeoxynucleotidesofc-mplinhibitedmegakaryocytecolonyformation.Theligandforc-mpl,thrombopoietin,wasclonedin1994.Thrombopoietinwasshowntobethemajorregulatorofmegakaryocytopoiesisandplateletformation.Theproteinencodedbythec-mplgene,CD110,isa635aatransmembranedomain,withtwoextracellularcytokinereceptordomainsandtwointracellularcytokinereceptorboxmotifs.TPO-Rdeficientmicewereseverelythrombocytopenic,emphasizingtheimportantroleofCD110andthrombopoietininmegakaryocyteandplateletformation.UponbindingofthrombopoietinCD110isdimerizedandtheJAKfamilyofnon-receptortyrosinekinases,aswellastheSTATfamily,theMAPKfamily,theadaptorproteinShcandthereceptorsthemselvesbecometyrosinephosphorylated.
