Secretoryandtransmembraneproteinsaresynthesizedonpolysomesandtranslocateintotheendoplasmicreticulum(ER)wheretheyareoftenmodifiedbytheformationofdisulfidebonds,amino-linkedglycosylationandfolding.Tohelpproteinsfoldproperly,theERcontainsapoolofmolecularchaperonesincludingcalnexin.CalnexinwasfirstidentifiedasbeinginvolvedintheassemblyofmurineclassIhistocompatibilitymolecules(1,2).Calnexinisacalcium-bindingproteinembeddedintheERmembranethatretainsthenewlysynthesizedglycoproteinsinsidetheERtoensureproperfoldingandqualitycontrol(3-5).Thespecificityofcalnexinforasubsetofglycoproteinsisdefinedbyalectinsite,whichbindsanearlyoligosaccharideintermediateonthefoldingglycoprotein(5).
